I don't believe he made it up. I think the term has been around for quite a while now. Perhaps some have heard it more with Prince than without. He almost certainly didn't make up the term "Vegas" to describe something that's glitzy and flash - I'm sure I've read critics and the like use the term "It was all a bit Vegas"....again, meaning a bit over-done. But, I haven't heard any other artist use the term as a musical direction, so I would suggest that its not a "musical" term per se. I think this is also known as a "vamp" although I'm not a musician. It means for the band to hold that chord until he counts off a change or ends it or moves on to something else. When Prince shouts "Vegas!" the band holds a major chord (usually the key the song is in, but he might call out a different key that will start the next song) with no particular rhythm, until Prince tells them to continue or stop. So the drummer would beat the shit out of that drumkit, the bassist would play whatever note it is, horns might go up and down the particular scale, etc.
Similar, but not the same (and less frequent) is when Prince shouts out "Confusion!", when the band does pretty much the same thing but in no particular key - resulting in a very chaotic sound. No idea if either of these are Prince's invention, or if he "borrowed" them from one of his heroes e.g. James Brown. "How embarrasing to be human!"
